Mechanism of action
Three proline substitutions, borrowed from rat amylin, disrupt the beta-sheet stacking that causes human amylin to form amyloid fibrils. The result is a soluble, formulable analogue retaining amylin receptor activity.

Why does native human amylin aggregate when rat amylin does not?
Differences in a small region of the sequence govern beta-sheet formation. Rat amylin carries prolines there, which break the sheet; human amylin does not, and forms amyloid. Pramlintide copies the rat prolines into the human sequence.
